CYTOKINE PRODUCTION BY WHOLE BLOOD CELLS AS AN INDEX OF THE INTENSITYOF THE POSTVACCINAL CELL-MEDIATED IMMUNITY

The possibility of evaluating the intensity of the cell-mediated immunity of healthy donors was investigated by estimation of cytokine production by whole blood cells in response to stimulation with HBsAg. Broad range of cytokines was measured after in vitro stimulation with PHA and HBsAg of whole blood of 48 healthy immunized by recombinant vaccine against hepatitis B and non-immunized volunteers. For the quantitative evaluation of the intensity of cellular immunity, the cytokine index Icyt (the ratio of antigen-stimulated to the mitogen-stimulated production of a cytokine) was used. It was shown that the values of cytokine indices for IFNγ, IL-6, IL-1β, TNFα, IL-1Ra were reliably differed in HBsAg vaccinated and non-vaccinated volunteers. The most significant difference between the volunteers and the greatest spreads of Icyt mean values were revealed in the index of interferon gamma (IIFNγ). There was no direct correlation between the levels of antibodies to HBsAg and IIFNγ (and absolute IFNγ values). The correlation between IIFNγ and IIL-6, ITNFα, IIL-1β, IIL-1Ra, allows assuming that the production of antibodies (driven by IL-6, IL-1β) as well as effector cells (regulated by TNFα and IL-1β) will be activated in persons with high IIFNγ under the contact with HBsAg.
