GENE EXPRESSION OF THE ENDOGENOUS BIOREGULATORS - CYTOKINES AND NEUROPEPTIDE OREXIN - UNDER STRESS CONDITIONS AND WITH ADMINISTRATION OF THE IMMUNOMODULATOR DERINAT

The present study was aimed to evaluate IL-1β, IL-10 (pro-inflammatory and anti-inflammatory cytokines respectively) and preproorexin mRNA expression in rats under experimental stress conditions. Two models of acute stress were used: cold stress (10 min cooling at -200‘ in individual cages) and combined stress (30 min cooling at -200‘ in combination with immobilization down on the back). The intensity of mRNA expression has been evaluated using real-time RT-PCR in splenocytes (for cytokines) and hypothalamus cells (for preproorexin). The results indicate that acute combined stress inhibits IL-1β mRNA expression and stimulates IL-10 mRNA expression in splenocytes. The possibility of correction of stress-induced alterations in gene expression of these bioregulators with immunomodulator Derinat has also been studied. Derinat abolishes stress-induced stimulation of IL-1β and preproorexin mRNA expression and stimulates IL-10 mRNA expression. (Cytokines and Inflammation. 2009. Vol. 8, No. 1. P. 18-22.)
