Changes of immunophenotype of murine dendritic cells during the maturation induction by proteins OprF and aTox of Pseudomonas aeruginosa.

Dendritic cells (DC) is a part of innate immune system, first element of immune response activation, including response to vaccine antigens. Vaccine against Pseudomonas aeruginosa, being developed in Mechnikov Institute, contains recombinant proteins OprF (outer membrane protein) and aTox (atoxic deletioned form of exotoxin A). Objective. To study the influence of recombinant proteins OprF и aTox of P. aeruginosa on the maturation of murine DC (immunophenotype changing). Materials and methods. Preparation: 25 μg of OprF, 50 μg of aTox sorbed on 75 μg of aluminum hydroxide, diluted in 0.5 ml PBS. DC were obtained from bone marrow cells of BALB/c mice by culturing with recombinant GM-CSF and IL-4 (Biosource, USA) for 6 days. Results. Test preparations caused a decrease in the number of immature cells, an increase in the cell population with adhesion markers, markers of antigenic presentation, costimulatory molecules and a terminal differentiation molecules. Proteins in the presence of adjuvant stimulated differentiation of DCs more actively; both myeloid and plasmacytoid DC were formed. Conclusions. The candidate Pseudomonas aeruginosa vaccine based on its recombinant OprF and aTox proteins induces the maturation of the DC generated from mice bone marrow cells. (Cytokines and Inflammation. 2017.
