CYTOTOXIC ACTIVITY OF IMMUNOGLOBULINS ISOLATED FROM BLOOD PLASMA OF HEALTHY DONORS AND MULTIPLE SCLEROSIS PATIENTS

The effects of immunoglobulin (Ig) preparations isolated from blood plasma of healthy donors and patients diagnosed for mild and severe forms of multiple sclerosis (MS), towards cultured leukemic cells of human (Jurkat, MT-4 and Namalwa lines) and mice (L1210 line) were compared. It was found that Ig of healthy donors possessed high cytotoxic activity (index of cytotoxicity IC " 50) towards all types of studied cells. Preparations of Ig of MS patients differentially affected T and B leukemic cells. They possessed cytotoxicity (IC " 30) towards B leukemic cells (Namalwa and L1210 lines), whereas their cytotoxicity towards T leukemic cells (Jurkat and MT-4 lines) in some cases was absent, and, oppositely, they stimulated proliferation of T cells. Study of the mechanism of cytotoxic activity of Ig by using microgelelectrophoresis of individual cells (DNA comet assay) showed that used Ig caused nuclear DNA degradation characteristic of apoptosis. Thus, the opposite effect of the autological antibodies of some MS patients on lymphocyte survival suggests an existence of considerable differences in the mechanisms of MS pathogenesis in various patients.
