WOUND HEALING AND LOCAL IMMUNOSTIMULATORY ACTIVITY OF RECOMBINANT HUMAN IL-1β IN PATIENTS WITH NON-HEALING WOUNDS AND TROPHIC LEG ULCERS

The aim of the study was to investigate the therapeutic effects of recombinant human IL-1β local application in patients with chronic wounds. IL-1β was used in the doses of 0.05 - 5000 ng/ml to treat non-healing wounds and trophic ulcers in patients with venous diseases or diabetes. Clinical assessment, wound areas measurements, cytological analysis and wound neutrophil functions (NBT-test and phagocytosis) study were performed before and several times during the treatment period. Treatment with IL-1β resulted in clinical improvement of wound healing in all groups of patients. Based on clinical results, the optimal concentration of IL-1β - 50 ng/ml - was chosen. IL-1β application caused a significant increase in number of wound leukocytes and epithelial cells as well as percent of macrophages and fibroblasts. IL-1β treatment stimulated spontaneous superoxide production and phagocytosis of wound neutrophils. Mechanisms of action of the preparation were the same in different groups of patients. Thus, IL-1β caused significant stimulation of wound reparation processes as well as activation of wound phagocyte functions which provides acceleration of wound healing, quicker clearance of wound areas and protects them from secondary infections.
