IMMUNOLOGICAL CONSEQUENCES OF ANTI-IFN GAMMA AND ANTI-IL-4 ANTIBODIES INDUCTION IN MICE

Certain forms of immune response were evaluated in mice having serum antibodies against key Th1 and Th2 cytokines IFN gamma and IL-4, induced by peptide immunization. The increase of spontaneous and mitogen-induced proliferation of splenic lymphocytes was found in mice immunized against IFN gamma. The ability of CD4+ T cells to differentiate into Th1 and Th2 did not change, but the autologous serum containing anti-IFN gamma or anti-IL-4 antibodies added to splenocyte culture suppressed differentiation of T helpers producing corresponding cytokines. Uptake of anti-IFN gamma antibodies leads to the suppression of both cellular and humoral immune response to ram erythrocytes, but does not influence anaphylactic antibody synthesis after immunization with ovalbumin. Uptake of anti-IL-4 antibodies suppresses humoral, but not cellular response to ram erythrocytes. Inhibitory influence of anti-IL-4 antibodies upon allergic response becomes clear when the anaphylactic antibodies are analyzed separately in mice with the high and the low anti-IL-4 antibodies levels (Cytokines and Inflammation. 2008.
