ACTIVATION OF HUMAN BLOOD BASOPHILES BY THE FCγ RECEPTOR LIGANDS: A CHOLINERGIC TUNING

A possibility of human blood basophiles activation by the Fcγ receptor ligands and the impact of the basophiles' cholinergic apparatus on histamine release were studied. The investigation was carried with the enriched basophiles fraction. Different Fcγ receptor ligands were used for basophile stimulation and different cholinergic agonists and antagonists (carbacholine, armin, methacin, hexametonium) were applied to influence the cholinergic cellular apparatus. Histamine release was measured with the Schore method. Membrane antigen CD203c (a marker of basophile activation) expression was assessed with flow cytofluorimetry. It was shown that all studied Fcγ receptor ligands (aggregated IgG, C-reactive protein, anti-CD16 antibody) activated histamine secretion in basophils. Cholinergic stimulation of basophiles also increased histamine secretion in dependence upon muscarinic and nicotinic type receptor activity.
