Effect of pentoxyphylline on Th2 cytokine production in women with infertility associated with minimal endometriosis

The aim of the work was to investigate in vitro and in vivo the influence of pentoxyphylline upon Th2 cytokine production and estimate the possibility of using pentoxyphylline in infertility associated with minimal endometriosis treatment. The immunological investigation of 70 women with infertility and endometriosis before and after treatment with pentoxyphylline in early postoperative period was carried out. The enriched population of lymphocytes was incubated in RPMI 1640 medium in the presence of pentoxyphylline for 24 h in 37°С and 5 % СО2 to elucidate it’s immunomodulatory effect in vitro. Intracellular cytokines were detected with monoclonal antibodies by two-color flow cytometry. It was found that the level of IL-4+ and IL-6+ cells within the lymphocyte and T-helper populations and the number of IL-5+ lymphocytes were significantly increased in infertile women with minimal endometriosis. Incubation of lymphocytes with pentoxyphylline in vitro led to the decrease of IL-6 production by lymphocytes of healthy women and diminishment of IL-4 and IL-6 production by lymphocytes and CD4+ cells of infertile women with endometriosis. After course of treatment by pentoxyphylline, normalization of intracellular cytokines production was observed in women with endometriosis. Immunomodulatory effect of pentoxyphylline in vitro was associated with beneficial clinical effect of the drug upon reproductive function of infertile patients with endometriosis.
